Clinico-pathological studies on the protective effect of some plant extracts on toxicity of some immunosuppressive drugs in rat

Master Abstract

Methotrexate (MTX) is a folic acid antagonist used in the management of autoimmune diseases, cancer, rheumatoid arthritis, and other inflammatory diseases through inhibition of the dihydrofolate reductase enzyme. However, its clinical use has been associated with a number of adverse effects and toxicities. The present study was conducted to evaluate the protective effect of the Lepidium sativum (LS) triterpene rich fraction and the Bougainvillea glabra (BG) flavonoids rich fraction on clinico -pathological alterations in rats intoxicated with MTX. One hundred and twenty male Wistar albino rats were divided equally into 6 groups. The first group was the control group (C). The second group (LS) was treated with the LS fraction only at a dose of 300 mg/kg b.w for one month. Rats in the third group (BG) treated with BG fraction at a dose of 200 mg/kg b.w for one month. The fourth group (MTX) was injected with methotrexate at a dose of 10 mg/kg i.p weekly for one month. The fifth group (LS+MTX) was treated with MTX and LS fraction as previously mentioned doses, and the sixth one was treated with MTX and BG fraction as previously mentioned doses. MTX injection induced a significant decrease in PCV%, Hb concentration, RBCs count, and WBCs count accompanied by lymphopenia and neutropenia. Besides elevations of urea, creatinine, TG, TC, LDL-c, VLDL, and AchE, a significant decrease in protein profile and HDL-c as well as alterations in oxidant/antioxidant markers were also observed. In addition, there was a significant downregulation of Nrf2, IL-10, and Bcl-2 genes with a significant upregulation of NF-kB, TNF-a, and caspase-3 genes. Significant histopathological alterations in the kidney, brain, and small intestine have also been observed. Co-administration of Lepidium sativum or Bougainvillea glabra fraction has a positive impact, as they substantially decreased the degree of MTX-induced pathological alterations.
