Some pharmacodynamic studies of Thymus vulgaris plant

Master Abstract

The Thymus vulgaris plant is commonly used in folk medicine since ancient times and is still used in the modern era. This has encouraged us to use this plant in the field of scientific research to investigate some phytochemical and toxicological as well as some pharmacological aspects of ethanolic extract of Thymus vulgaris. Thyme leaves has been bought from the Faculty of Pharmacy, Cairo University, it was dried and the preparation of the ethanolic extract was obtained. The preliminary phytochemical screening that carried out on the plant indicated the presence of alkaloids, carbohydrates and glycosides, flavenoids, resins, saponins, tannins and unsaturated sterols and triterpenes. The result of acute toxicity study (LD50) of ethanolic extract of Thymus vulgaris showed absence of toxicological symptoms, mortality and post-mortem changes up to oral administration of 7000 mg/kg body weight. Concerning the pharmacological studies, pretreatment of rats with ethanolic extract of Thymus vulgaris at dose levels of 250 and 500 mg/kg body weight protects the gastric mucosa against damaging effect of ethanol and exerts antiulcerogenic effect. Topical application of ethanolic extract of Thymus vulgaris in the form of ointment (10 %) on the induced wounds of rats posses healing activity so it can be used in treatments of fresh wounds. Summary 143 Oral administrations of ethanolic extract of Thymus vulgaris posses analgesic, anti-pyretic and anti-inflammatory activities, thus; it is considered as a good source in treatment of abscesses, fever, inflammations and different types of pain. Furthermore, it significantly prolonged thiopental sodium sleeping time in mice at both doses (250 and 500 mg/kg body weight). The prolonged administration of ethanolic extract of Thymus vulgaris at dose levels of 250 and 500 mg/kg body weight daily for two months improved the body weight and showed no significant changes in blood picture during and after stopping administration. Also, there was significant increase in total leucocytic count in case of high dose only (500 mg/kg body weight) during four weeks of administration without any significant changes in differential leucocytic count. There were no significant changes in liver and kidney functions during four weeks of administration and after stopping of administration. During eight weeks of administration, there was significant decrease in urea and creatinine levels. Ethanolic extract of Thymus vulgaris resulted in inhibitory effect in force of intestinal contractility of rabbits starting from slight inhibition in force of contraction in concentration of 0.05 mg/ml bath and ending with complete relaxation in concentration of 3 mg/ml bath. The site of action was proved to be atropine-like. Therefore, it can be used in colicy pain. The effect of ethanolic extract of Thymus vulgaris on uterine motility in cases of non pregnancy (estrus and non estrus stages) and pregnancy was found to be inhibitory effect in force starting from 0.05 mg/ml bath and ending with complete relaxation in concentration of 2 Summary 144 mg/ml bath in case of estrus stage and 4 mg/ml bath in case of pregnancy stage. So, it can be used in cases of threatened abortion. It also has antibacterial effect against E. coli O78 and Bacillus subtilis ATCC 25922. Therefore, it is recommended to be used as a natural antimicrobial agent without causing health hazard as drug tolerance.
